# Semax: research reference

For research use only. Checked November 1, 2026.

Semax is a synthetic peptide described in primary papers as ACTH(4–7)PGP, with the sequence Met–Glu–His–Phe–Pro–Gly–Pro. Selected studies examine transcriptional and protein responses after experimentally induced cerebral ischemia in rats. These studies can inform mechanistic questions, but they do not establish that a retail material has the same identity or that a molecular change predicts a clinical outcome.

## Identity
The sequence and exact terminal chemistry matter more than a marketing name. This reference concerns the Semax material described by the cited investigators, not automatically every acetylated, amidated or otherwise modified product sharing the name. Require an identity specification before applying the literature to a sample.

## Mechanism
A 2014 rat focal-ischemia study linked Semax exposure with changes in immune- and vascular-associated transcripts. This is a gene-expression observation in injured tissue, rather than demonstration of one exclusive receptor mechanism. [semax-2014]

A later ischemia-reperfusion experiment examined selected proteins including CREB, JNK, MMP-9 and c-Fos. Different tissue regions showed different patterns, illustrating why a single pathway slogan loses important experimental context. [semax-2021]

Read the model, tissue and sampling time together. A transcript increase, a protein change and a behavioral endpoint are different measurements; agreement should be demonstrated rather than assumed.

## Selected research
### Transcription after focal ischemia
Model: Rat brain cortex after permanent middle cerebral artery occlusion

Investigators observed changes in genes associated with immune activity and vascular processes in the Semax-exposed groups.

Limitation: The findings concern experimentally injured rat tissue and selected observation times; they are not a general cognitive-performance result.

### Selected protein responses
Model: Rat transient ischemia-reperfusion model

Protein profiling reported region-specific changes in markers associated with inflammation, cell-death signaling and recovery pathways.

Limitation: The selected marker panel does not establish a complete mechanism or predict outcomes in an unrelated experimental system.

## Open questions
- Which observed molecular changes are necessary causes rather than downstream correlates?
- How consistently do independent laboratories reproduce the findings across model, time point and material specification?
- How much do terminal modifications or formulation differences change experimental behavior?
- This selected-source summary is not a systematic review or a complete inventory of clinical evidence.

## Storage and handling
The cited mechanistic papers do not establish a universal commercial shelf life. Keep the storage instruction for the exact material separate from a biological study result.

Record sequence/terminal specification, salt or counterion, supplied form, lot, certificate, packaging and documented conditions. Do not borrow solution stability from a different formulation.
